When you have achieved the indicated requirement the corresponding Signature Konami character will become unlocked.

Unlock Evil Rose:
Beat Medium Career.

Unlock Pentarou:
Beat Hard Career.

Unlock Sparkster:
Beat Easy Career with ALL Gold medals.

Unlock Simon Belmont:

Unlock Solid Snake:
